Abstract

Larutan Turk merupakan bahan pemeriksaan leukosit manual dengan komposisi asam asetat glasial, gentian violet dan aquadest. Asam asetat glasial dalam larutan Turk merupakan asam lemah dengan pH 2,4. Jeruk nipis (Citrus. aurantifolia S.) memiliki kandungan asam sitrat yang juga merupakan asam lemah. Asam lemah dapat melisikan sel darah selain leukosit yang berguna untuk memudahkan pemeriksaan hitung jumlah leukosit manual. Tujuan dari peneliti adalah untuk memberikan gambaran perbandingan jumlah leukosit yang dihitung menggunakan modifikasi air perasan jeruk nipis (C. aurantifolia S.) dengan konsentrasi 2%, 3%, 4% dan 5%. Design penelitian berupa deskriptif. Populasi berupa 1 orang Mahasiswa Analis A angkatan 2017 STIKes ICMe Jombang sejumlah 1 darah Mahasiswa. Sampling menggunakan total sampling. Variabel adalah Modifikasi air perasan jeruk nipis (C. aurantifolia S.). Alat ukur berupa observasi laboratorium. Pengolahan data dengan editing, coding dan tabulating. Analisa data berupa analisis deskriptif. Hasil penelitian ini, didapatkan hasil 10.900 sel/mm3 larutan Turk sebagai kontrol, sedangkan modifikasi air perasan jeruk nipis (C. aurantifolia S.) dengan konsentrasi 2% diperoleh 11.900 sel/mm3, konsentrasi 3% diperoleh 8.550 sel/mm3, konsentrasi 4% diperoleh 8.000 sel.mm3, dan konsentrasi 5% diperoleh 7.900 sel/mm3. Sehingga konsentrasi 2% merupakan konsentrasi paling efektif dengan perbandingan hasil yang mendekati jumlah leukosit pada larutan kontrol. Kesimpulan modifikasi air perasan jeruk nipis (C. aurantifolia S.) dapat digunakan sebagai alternatif pengganti komposisi larutan Turk dalam pemeriksaan hitung jumlah leukosit paling efektif dengan konsentrasi 2%.
